Quantification of inorganic as well as organic phosphates (such asphospholipids, phosphoproteins, and nucleid acids) is of interest in many biological studies.Phosphatase activity, for instance, can be derived from such an analysis after co-incubationof a phosphatase and a specific phosphopeptide substrate (J. Cell. Biochem. 77:445,2000).Similarly, an assay for phospholipase activity can be performed after hydrolysis ofphospholipids by phospholipases. Although various methods can be employed to measurephosphates, non-radioactive colorimetric protocols suitable for high-throughput analysis arethe methods of choice in most laboratories. Perhaps the most sensitive colorimetric methodfor phosphate assay is based on complex formation of phosphomolybdate with the basic dyeMalachite Green. Our Phosphate Assay Kit(sufficient for 400 assays) is formulated to maximize the sensitivity and stability of thedye-phosphomolybdate reaction. The kit is stable for at least one year if handled and storedproperly. The assay, compatible with microplate formats, is simple and can be finished in 15 -20 min. Reaction product is quantified by absorbance at 600-660 nm.
